L-arginine is absorbed directly, though a portion is broken down by arginase in the gut wall and liver on the way. L-citrulline largely escapes that breakdown and is converted to arginine in the kidneys, arriving later and lasting longer.
In the endothelium — the single-cell layer lining every blood vessel — the enzyme nitric oxide synthase converts arginine into nitric oxide and citrulline. The citrulline produced can be recycled back into arginine, which is why the two amino acids form a loop rather than a one-way path.
